According to our proposed working program we analyzed the requirement of Ybx1 for development of Jak2^V617F driven disease as well as for maintenance and cell cycle activity of MPN stem cells. To dissect the requirement of Ybx1 for disease progression and maintenance of MPN stem cells we used genetically engineered mouse models to achieve conditional deletion of Ybx1 and conditional activation of the Jak2^V617F mutation. It was also our aim to elucidate mechanistic consequences of Ybx1 inactivation. We analyzed signaling pathways downstream of Jak2/EpoR to determine effects on major signaling nodes that may be induced by activation/deletion of Ybx1.

The most frequent cause of therapy resistance is point mutations in the BCR-ABL tyrosine kinase domain. Preliminary studies of the research group identified a high prevalence of BCR-ABL mutant subclones that cannot be detected by conventional analytical methods (e.g. Sanger sequencing). However, the clinical significance of these subclones is still unclear. In the proposed research project, the prevalence, kinetics and prognostic significance of BCR-ABL mutant subclones will be systematically investigated in a clinically well-characterized patient cohort using a highly sensitive next-generation sequencing method.

Prevention concept for young people after cancer

Long-term survivors of childhood, adolescent or young adult cancer (CAYA) are at increased risk for disease- or therapy-related long-term effects. To address the special needs of this patient population and provide individualized follow-up care, the CARE-for-CAYA program was established. The program evaluated whether needs-adapted interventions in the areas of sport, nutrition and psycho-oncology lead to an improvement in lifestyle and/or psychosocial well-being.

Against the backdrop of an increasing application of magnetic hybrid materials in biomedicine, a better understanding of the processes involved in the interaction of these hybrid materials with cells is becoming more and more important. Therefore, the following aspects should be examined in this project in detail :

1. shape, charge and chemical composition of the magnetic hybrid materials and their influence on the subsequent processes,
2. the biological identity that the materials acquire by the essential contact with biological fluids and which is represented in particular by the protein corona,
3. the contact with the cell surface and the resulting consequences for the nanomaterials, such as the relaxation behavior and the structuring of the cell membrane,
4. tracking the further handling of the cell with the nanomaterial and
5. the consequences of these processes for the use of magnetic hybrid materials in the context of therapeutic measures, especially in pregnant women and in cancer therapy.
