LSM880 with AiryScan for high-resolution microscopy

The LSM880 with AiryScan and 34-channel spectral detector, together with the laser equipment, forms the basis for research projects in the field of high-resolution microscopy. The focus of the use of the system is on the investigations of the structure of the endoplasmic reticulum (ER) and the dynamics of the ER and on the effects of structural ER changes on different ER functions in selected diseases. In addition to projects on neurodegenerative diseases, the role of the ER in the development of cancer cells, the effects of a reticulopathy disorder in sepsis, and the role of another ER function, namely lipid synthesis, in cardiomyocytes in the development of heart failure will be investigated ,

All of these research topics require an increase in optical resolution of a standard laser scanning microscope. The structures of the ER are beyond 220 nm, which is the resolution limit of a standard confocal laser scanning microscope at 488 nm wavelength and using a 63x / 1.4 objective. Also needed is a quantitative fluorescence microscopy system capable of simultaneously detecting the entire usable spectral range (400-800 nm) to analyze emission spectra for overlap and autofluorescence and to identify multiple stains by unmixing.

The funding will be provided by the European Regional Development Fund (ERDF OP 2014-2020) and from the national funds.
